Although the Th1/Th2 paradigm is generally applicable to allergy and autoimmunity, in mucosal immunity, in particular in the gastrointestinal tract, a different paradigm is beginning to be recognized. In the gut mucosa, regulatory T-cells (Treg) play an important role for maintaining intestinal homeostasis by suppressing the excessive immune response associated with enteric pathogens and allergens. Intestinal Treg are characterized by the expression of CD4 and the forkhead/winged helix transcription factor (FOXP3), and are induced by the products of gut commensal bacteria. Treg act by releasing anti-inflammatory cytokines, such as IL-10 and transforming growth factor (TGF)-beta, which prevent excessive inflammation in the gut and suppress the generation of immunogenic effector T-cells. Treg can be divided into peripheral Treg in the circulation and mucosal Treg that are localized in the gut mucosa. It has been recently demonstrated that both peripheral and mucosal Treg induce the differentiation of non-Treg into Treg through cell-to-cell contact and through secretion of cytokines. Here, I summarize recent findings on the immune system and regulatory mechanisms in the gut and discuss the roles of the two types of Treg for intestinal homeostasis.Q: k-means cluster: how many clusters should be returned? When I do k-means clustering, is it better to return a reasonable number of clusters, or is a more "correct" number of clusters better? How does one get to a "correct" number? A: This is more of a maths question and probably not the right place to ask it,
